Distinct from MCED (BIO-CFDNA-METHYLATION-MCED) in that ctDNA-MRD assumes a known prior cancer diagnosis with post-curative-intent surgery / chemoradiation. Strongest evidence in CRC (GALAXY / CIRCULATE-Japan, Kotani Nat Med 2023): MRD-positive post-op patients ~22% 18-mo DFS vs ~93% MRD- negative; MRD-positive benefit from adjuvant chemotherapy. Emerging evidence in muscle-invasive bladder cancer (IMvigor010 ctDNA subgroup), early breast cancer (c-TRAK-TN), NSCLC (post-IMpower010 retrospectives), pancreatic, gastric.
