Notes
MOUNTAINEER Phase II (2022): HER2-amp RAS-WT chemo-refractory mCRC, ORR 38%, mPFS 8.2 mo, mOS 24.1 mo. FDA-approved 2023 for HER2+ RAS-WT mCRC (chemo-refractory). Tucatinib is a HER2-selective TKI with low EGFR cross-inhibition — favorable diarrhea profile vs lapatinib. Trastuzumab moiety provides cardiotoxicity surveillance requirement (baseline + q3 mo LVEF). Chemo-free regimen advantage vs T-DXd.
