KRYSTAL-1: ORR 43%, mPFS 6.5 mo, mOS 12.6 mo in pretreated KRAS G12C+ NSCLC. Intracranial ORR 33% — preferred over sotorasib for active brain metastases. Dosing TID (3x200 mg) sometimes used to improve GI tolerability. KRYSTAL-12 phase-3 vs docetaxel ongoing.
