TROPION-Lung01 (Ahn 2023) phase-3 randomized previously-treated metastatic NSCLC to Dato-DXd vs docetaxel (mPFS 4.4 vs 3.7 mo, HR 0.75; benefit concentrated in non-squamous subgroup mPFS 5.6 vs 3.7 mo, HR 0.63). TROP2 is broadly expressed across NSCLC histologies (IHC enrollment not required) — TROP2 IHC is informational, not gating. Candidate RF positions Dato-DXd in 2L+ non-squamous NSCLC as a non-platinum cytotoxic alternative to docetaxel.
