Notes

TAGS trial (Shitara 2018): 3L+ gastric/GEJ adenocarcinoma after ≥2 prior regimens — TAS-102 vs placebo mOS 5.7 vs 3.6 mo (HR 0.69), PFS 2.0 vs 1.8 mo. NCCN cat 1 for 3L+ gastric. Distinct mechanism from prior fluoropyrimidine exposure (TS-independent thymidine analog incorporation) — active in 5-FU-refractory disease. Main toxicity is myelosuppression (neutropenia ~38%). Hand-foot reactions notably less than fluoropyrimidine.
