Notes

Phase III trial (Demetri 2006 Lancet): sunitinib vs placebo in imatinib- resistant/intolerant GIST — mPFS 27.3 vs 6.4 weeks (HR 0.33, p<0.0001). Trial stopped early. FDA approved 2006. KIT exon 9 or wild-type GIST may derive greater benefit than exon 11. Continuous 37.5 mg dosing is an alternative to the 4/2 schedule (per some studies, similar efficacy, improved tolerability). Regorafenib is standard 3L after sunitinib failure.
