Notes
Pan-BCR-ABL1 TKI active against ALL kinase-domain mutations including T315I gatekeeper. PACE trial: 56% MCyR / 34% MMR in CML-CP. Reserved for T315I+ OR post ≥2 TKI failure due to substantial vascular toxicity. Response-adjusted dosing per OPTIC reduces vascular events. Asciminib (STAMP) is emerging less-toxic alternative for non-T315I post-multi-TKI; ponatinib retains advantage at standard dose for T315I (head-to-head asciminib at 200 mg BID for T315I emerging). STUB — pending Co-Lead sign-off.
