Notes
COLUMBUS phase 3: ENCO 450 + BINI 45 BID vs vemurafenib in 1L/2L BRAF V600-mutant melanoma — mPFS 14.9 vs 7.3 mo (HR 0.54); 5-y OS 33.6 vs 16.9 mo. Tolerability advantage over vemurafenib (less photosensitivity) and dabrafenib+trametinib (less pyrexia, less arthralgia). Position: NCCN/ESMO preferred BRAFi+MEKi doublet for BRAF V600E/K metastatic melanoma. In our 2L+ algorithm: used for BRAF V600+ patients who progressed on 1L IO. dabrafenib + trametinib (REG-DABRAFENIB-TRAMETINIB-NSCLC) is an interchangeable alternative — both NCCN cat 1.
