ZUMA-2 trial (Wang 2020): r/r MCL after prior cBTKi (median 3 prior lines, ~25% TP53-mut, 60% high-risk MIPI) — ORR 91%, CR 68%, 3-year OS ~60%. First CAR-T for MCL. Higher Grade ≥3 ICANS in MCL cohort (~31%) vs DLBCL — MCL patients older + heavily pretreated. CD19-depletion manufacturing step in brexu-cel reduces malignant apheresis contamination (vs axi-cel) — relevant in leukemic MCL. Bridging therapy critical: TP53-mut / blastoid MCL progresses fast in 3-4 wk manufacturing window; pirtobrutinib bridging preferred if not prior-failed.
