Notes
ESCAT IA. EMBRACA established talazoparib (1 mg PO daily) for germline BRCA1/2-mutant HER2- metastatic breast (≤3 prior cytotoxic regimens). Comparable efficacy to olaparib (no head-to-head trial), different AE profile: more haematologic (anaemia G3+ ~39%), less GI than olaparib. Patient preference and AE-profile drive the olaparib-vs-talazoparib choice. KNOWN GAP: dedicated REG-TALAZOPARIB-BREAST regimen entry not yet authored — current placeholder points to REG-OLAPARIB-BREAST. Engine trace must surface that talazoparib dosing differs (1 mg PO daily monotherapy continuous; reduce to 0.75 mg if anaemia, then 0.5 mg). Follow-up authoring task. UA access: talazoparib not reimbursed.
